Sermorelin is the synthetic form of GHRH(1-29), the first 29 amino acids of human growth hormone-releasing hormone. It is the original GH-releasing peptide — the first GHRH analog to be developed, clinically tested, and approved for therapeutic use (for pediatric growth hormone deficiency). While newer analogs like CJC-1295 and Tesamorelin have improved upon its pharmacokinetics, Sermorelin remains the most extensively studied GHRH peptide in the research literature, with decades of clinical data spanning pediatric, adult, and geriatric populations.
Sermorelin binds to the GHRH receptor on anterior pituitary somatotrophs, activating the adenylyl cyclase-cAMP-PKA pathway to trigger GH release. As the unmodified (1-29) fragment, it is rapidly degraded by serum proteases, giving it a plasma half-life of 10-20 minutes. This short half-life means Sermorelin produces acute, physiological GH pulses that closely mimic the body's endogenous secretion pattern. This is considered advantageous by researchers who prioritize the natural pulsatile rhythm over maximized GH output — the natural pattern preserves GH receptor sensitivity and negative feedback integrity more effectively than sustained-release analogs.
Research spanning several decades documents Sermorelin's effects on GH secretion, IGF-1 elevation, body composition, sleep architecture, and functional markers of aging. Clinical studies in adults demonstrated increases in lean body mass, reductions in body fat percentage, improvements in skin thickness and texture, enhanced deep (slow-wave) sleep, and improvements in exercise recovery. Notably, Sermorelin's long safety record has made it a reference compound against which newer GHRH analogs are benchmarked. Studies show it maintains efficacy without tachyphylaxis (tolerance) over extended use, as it works through the body's own GH-releasing machinery rather than bypassing it.

Reconstitute with 2.5ml bacteriostatic water (yielding 2mg/ml). Administer via subcutaneous injection, typically 100-300mcg per dose, once daily before bedtime. The pre-sleep timing aligns with the body's natural nocturnal GH pulse. Sermorelin should be administered on an empty stomach (at least 2 hours after last meal). Due to its short half-life, consistency in daily administration is important for sustained IGF-1 elevation. Store at 2-8C after reconstitution. The 5mg vial supports 17-50 days of research at standard dosing.
Sermorelin is supplied as a lyophilized (freeze-dried) powder and must be reconstituted with bacteriostatic water (BAC water) before use in a research setting.
- Clean the BAC water vial stopper and the peptide vial stopper with an alcohol swab. Allow to dry.
- Draw the required volume of BAC water into a sterile syringe (typically 1–3 mL depending on target concentration).
- Angle the needle so the water runs down the inside wall of the peptide vial. Avoid dispensing directly onto the powder.
- Do not shake. Gently swirl or roll until fully dissolved. Vigorous shaking can denature peptides.
- Refrigerate reconstituted solution at 2–8°C. Most reconstituted peptides are stable 14–30 days depending on compound.
